Acute Porphyria Drug Database

C03CC01 - Etacrynic Acid
Propably not porphyrinogenic
PNP

Rationale
CAVE: under revision. Water-soluble substance. Non-CYP metabolism. No data pointing to CYP-interaction.
Chemical description
Etacrynic acid is not a sulfonamide congener unlike many other diuretics.
Therapeutic characteristics
Loop-diuretic, short acting.
Metabolism and pharmakokinetics
Conjugation occurs in the liver with compounds containing sulfhydryl groups, such as cysteine and N-acetylcysteine. This seems to be the major metabolic pathway. Etacrynic acid is excreted both in the bile and the urine, partly unchanged and partly in the form of metabolites. An in vitro study with rat liver microsomes indicates oxidative metabolism via the CYP450-system. It is not known to which extent such metabolism occurs in man.
Preclinical data
None. No data pointing to CYP-induction.
Personal communication
None
Published experience
